Spatial Multiomics

Spatial Multiomics - Instruments

  • Xenium (10x Genomics)

Overview: The Xenium platform and assays facilitate detection of in situ gene expression of hundreds to thousands of RNA molecules using detection probe sin formalin-fixed paraffin-embedded (FFPE) and fresh frozen (FF) tissue samples with cellular resolution.

Further information on the Xenium system, as well as assay options and protocol documentation can be found on the 10X Genomics website here:

  • CosMx (NanoString)

Overview: CosMx spatial molecular imager (SMI) enables high-plex detection of thousands of RNA molecules in formalin-fixed paraffin-embedded (FFPE) and fresh frozen (FF) tissue samples with cellular resolution. CosMx additionally allow for imaging of up to 64 proteins (via antibody detection) but on a subsequent slide (i.e. RNA and protein detected on separate sections.

  • Visium/CytAssist (10x Genomics)

Overview: The Visium platform from 10x Genomics combines histology and spatially resolved whole transcriptome gene expression profiling to localize and quantify gene expression in the tissue context. It is based, in part, on the Spatial Transcriptomics methodology (1). Visium was commercialized in 2019 and has been used in groundbreaking papers demonstrating the breadth of its applications, including cancer (3,4), neuroscience (5), immunology (6), and developmental biology (7). Many researchers have adopted the assay, as evidenced by the over 400 peer-reviewed publications and pre-prints utilizing the technology. Visium Spatial Gene Expression is compatible with a variety of tissue types and species.
